In prefatory remarks to ten contributed chapters by scientists and clinicians involved in developing a new generation of drug therapies for treating cancer, Dr. Schwartz (Memorial Sloan-Kettering Cancer Center, New York, NY) introduces the promise of these approaches based on advances in understanding the molecular basis of the tumor cell cycle. He explains that because, unlike classic chemotherapy drugs and radiation, these agents are not cytotoxic, they are most effective in combination with traditional therapies. Agents discussed include proteins regulating the cell cycle (e.g., cyclin-dependent kinase inhibitors), and those targeting cell-surface receptors (e.g., EGF-R, HER2).
